Redox properties of a standardized extract of Chenopodium quinoa Willd fruit pericarp modify rat liver GST activities

    Get PDF
Research of antioxidant properties of herbal thiol compounds is scarce. The main non-enzymatic antioxidant compounds in animal cell are GSH and cysteine. Therefore, in this work, we studied the redox effects of a Chenopodium quinoa Willd (Quinoa) coats extract previously titrated in its polyphenol and thiol compounds. The effects of quinoa extract on rat liver cytosolic and microsomal GSH-transferase was tested. The catalytic active form of this enzyme is its disulphide dimer, and then its reduction provokes its inactivation. Quinoa extract inhibited both enzymatic activities in a concentration-dependent manner. The reducing power of this extract was significantly higher than N-acetyl-cysteine and dithiothreitol. Low concentrations of quinoa extract (without surfactant properties) decrease the apparent Vmax of both cytosolic and microsomal GST, increase their apparent GSH Km and not modify their apparent Km for substrate 1-chloro-2,4-dinitrobenzene. Redox effects of Quinoa extract would be the main cause involved in the inhibition of GSH-transferase activities. Moreover, thiol compounds present in this extract and not polyphenols seem to be the most important reducing agents acting on disulphide bond of GST active dimer. New pharmacological experiments are being carried out in order to evaluate the redox importance of thiol compounds present in this Quinoa extract.

PregradoQuímico(a) Farmacéutic

From September 1993 to August 2001, 7,190 phlebotomine were collected with CDC light trap in an endemic area for human leishmaniasis, in the departments of Misiones and Itapúa, Paraguay. Eleven species were identified: Lutzomyia neivai (93.7%), L. whitmani (4.1%), and L. fischeri, L. shannoni, L. migonei, L. misionensis, L. cortelezzii, L. pessoai, L. alphabetica, Brumptomyia avellari and B. guimaraesi (less than 1%). The last three species are new records for the country. The biodiversity and phlebotomine abundance were associated with the proximity to primary forest or gallery forest, but L. neivai was also found in peridomestic periurban environment. L. neivai was found throughout the year, and showed a period of higher activity from September to April (spring to fall) with a unimodal or bimodal pattern in relation to the annual rainy peaks during the summer. Background literature about phlebotomine from Paraguay has been reviewed.Fil: Salomón, Oscar Daniel. Ministerio de Salud de la Nación; ArgentinaFil: Rossi, Gustavo Carlos. Proteolytic cleavage of Arabidopsis thaliana phosphoenolpyruvate carboxykinase-1 modifies its allosteric regulation

Phosphoenolpyruvate carboxykinase (PEPCK) plays a crucial role in gluconeogenesis. In this work, we analyze the proteolysis of Arabidopsis thaliana PEPCK1 (AthPEPCK1) in germinating seedlings. We found that the amount of AthPEPCK1 protein peaks at 24-48 h post-imbibition. Concomitantly, we observed shorter versions of AthPEPCK1, putatively generated by metacaspase-9 (AthMC9). To study the impact of AthMC9 cleavage on the kinetic and regulatory properties of AthPEPCK1, we produced truncated mutants based on the reported AthMC9 cleavage sites. The Δ19 and Δ101 truncated mutants of AthPEPCK1 showed similar kinetic parameters and the same quaternary structure as the wild type. However, activation by malate and inhibition by glucose 6-phosphate were abolished in the Δ101 mutant. We propose that proteolysis of AthPEPCK1 in germinating seedlings operates as a mechanism to adapt the sensitivity to allosteric regulation during the sink-to-source transition.Fil: Rojas, Bruno Ezequiel. Training Design, Performance Analysis, and Talent Identification—A Systematic Review about the Most Relevant Variables through the Principal Component Analysis in Soccer, Basketball, and Rugby

    Get PDF
Since the accelerating development of technology applied to team sports and its subsequent high amount of information available, the need for data mining leads to the use of data reduction techniques such as Principal Component Analysis (PCA). This systematic review aims to identify determinant variables in soccer, basketball and rugby using exploratory factor analysis for, training design, performance analysis and talent identification. Three electronic databases (PubMed, Web of Science, SPORTDiscus) were systematically searched and 34 studies were finally included in the qualitative synthesis. Through PCA, data sets were reduced by 75.07%, and 3.9 ± 2.53 factors were retained that explained 80 ± 0.14% of the total variance. All team sports should be analyzed or trained based on the high level of aerobic capacity combined with adequate levels of power and strength to perform repeated high-intensity actions in a very short time, which differ between team sports. Accelerations and decelerations are mainly significant in soccer, jumps and landings are crucial in basketball, and impacts are primarily identified in rugby. Besides, from these team sports, primary information about different technical/tactical variables was extracted such as (a) soccer: occupied space, ball controls, passes, and shots; (b) basketball: throws, rebounds, and turnovers; or (c) rugby: possession game pace and team formation. Regarding talent identification, both anthropometrics and some physical capacity measures are relevant in soccer and basketball. Although overall, since these variables have been identified in different investigations, further studies should perform PCA on data sets that involve variables from different dimensions (technical, tactical, conditional)
